Wat is IL-2?

Interleukine 2 (IL-2) is een eiwit dat gemaakt wordt in kleine hoeveelheden door leukocytes (witte bloedcellen). Middels moleculair biologisch technieken kan het ook in grote hoeveelheden worden gemaakt. IL-2 heeft veel functies. De belangrijkste functie is het stimuleren van immuun (afweer) reacties (T lymfocyten, NK cellen). IL-2 is veroorzaakt ook vaatlekkage. Vaatlekkage is de belanrijkste complicatie van systemische IL-2 therapie, maar niet bij lokale IL-2 therapie.

Mechanisme van IL-2 therapie tegen kanker

IL-2 wordt gebruikt voor de behandeling van kanker. Oorspronkelijk werd IL-2 systemisch toegediend; dit heeft slechts een beperkte werkzaamheid en een hoge toxiteit (giftigheid). Wij kijken daarentegen naar lokale IL-2 therapie. Lokale toediening van IL-2 in de tumor geeft nauwelijks bijwerkingen en kan leiden tot gedeeltelijke of gehele tumor regressie en zelfs genezing.

Lokale IL-2 therapie van cancer

We focus on application of IL-2 therapy at the site of the tumour (local IL-2 therapy). Generally this leads to about 50-70% objective responses. Local IL-2 therapy is hardly toxic; it only causes local inflammation at the site of injection. We are studying the mechanism of tumour regression induced by local IL-2 application; both vascular leakage and immune stimulation seem to be involved. Histological studies show that local IL-2 application induces edema. This will cause reduced supply of nutrients and oxygen to the tumour cells. This increases necrosis. The dead cells will lead to an immune reaction, because cured mice and cured guinea pigs are specifically immune to the original tumour.
