More than a half of my stay in Japan has passed since I will be back to Korea on this December. I am accustumed to be here, but still poor in Japanese. The weather is so hot like in an oven. I could get some protoplasts of Tricholoma species, but still need to check the rate of regeneration. I am checking temperature and the concentration of enzymes. If the result shows good, then I will go further to make a fusant of those species. I am so anxious to have a distinctive result for the experiment but ...... |

I prepared protoplasts of Tricholoma matsutake (pine-mushroom fungus). At first, I have trained with Coprinus cinereus Fis., which was very easy to handle. But, it was so difficult to work with Tricholoma matsutake. Now, I made a basic scheme for the preparation of mycelial samples for protoplast preparation. I am trying to prepare the protoplasts of Tricholoma matsutake with high yield. I visited Fukuoka for attending an academic meeting of Japan Society for Bioscience, Biotechnology, and Agrochemistry(3/29-4/1). I also attended the meeting of Japanese Mycological Society from 5/20 to 5/23, and I could meet good scientist of mushrooms in Japan. I could get some idea about cell culture and fusion-related things. I also attended the third annual meeting of Japanese Applied Mushroom Science and Biotechnology from 7/15 to 16, and I presented a brief outline of Korean research for the concern of Tricholoma matsutake. |

As you may noticed from my personal records, I have three children with my wife. The first one is seven years old, and she is attending the Onan elementary school in Namiki, Tsukuba city. The second one is five years old, and she goes kindergarten which is managed by Japanese church, the Megumi(blessing in English) church, located in Tsuchiura city. The first one is in second grade, and the second daughter is the eledest in their kindergarten. The last one stays home since he is too young (just thirty one months old) to go somewhere. Thus, my wife is still busy to take care of the boy. Anyway, all the family are healthy and we are very happy to be here. They are in summer vacation and my wife is so busy to take care of those. |

My goal to accomplish here in Japan is to make a fusant of Tricholoma species. Thus, I am practicing a technique to prepare protoplast of Tricholoma species with a standard enzyme, now. But, I am still in trouble to prepare large amount of young mycelia of Tricholoma strains. Later, I will try to find suitable combination of enzymes, time and temperature of enzyme treatments and so on. Nowadays, I am getting busy as I know several people in this region. But, still in the condition of idle compared that in Korea.
