He married Patience Sprague. Patience was born in Providence, RI about 1675. She was the daughter of Jonathan Sprague and Mehitable Holbrook.

William Jenckes and Patience Sprague had the following child:

3 i. Ester³ Jenckes was born on (birth date unknown). She married John Comstock. He was the son of Samuel Comstock and Elizabeth Arnold. (See John Comstock for the continuation of this line.)
