The Basics

Creating a Schedule:

Once you know the number of teams in your league, you need to create a schedule.  The easiest way to do this is to buy one of the fantasy football books sold in a bookstore.  They all show a schedule based on the number of teams you have.  Most books show 8 or 10 team leagues with one or two divisions.  Choose the format that you like the best.  You also need to determine whether you want a playoff system at the end of the year or choose to have everyone play every game.  There are advantages and disadvantages to each method.

With Playoff:

This method rewards the top teams to get a bye when the playoffs start (similar to the NFL).  The lower seeds play each other in earlier rounds.  This system usually winds up play on the 15th game of the NFL season.  The reason for this is that some of the better teams usually rest their players the last week of the season prior to the playoffs.  Normally the better fantasy football performers are on the best teams.  This method can allow the team playing the best at the end of the year to win.  The disadvantage of this method is that by game 13 of the season, some of the teams are done playing.  Others give up earlier when they know they have no shot at the playoffs.

Without Playoff:

This method allows all teams to play up until the last week of the season.  Teams with bad records can still be a spoiler to determine who wins.  This method is survival of the fittest and the team consistently playing the best throughout the year will win.  The disadvantage to this method is that a team who drafted players that do not perform well early cannot make up enough ground to win.  In a playoff system, all he needs to do is make the playoffs and play week to week to win it all.
