Kaoru is the assistant instructor in the Kamiya Kasshin dojo.She first met Kenshin when she attacked him thinking that he was the one causing murders in Tokyo in the name of her sword style, because he was carrying a sword. In Meiji times, it was forbidden to carry a sword. After saving her from the person who is really causing the murders, Kaoru invites Kenshin to live at her dojo.

Serving as the "Manslayer in the Shadows" during the revolution for the Ishin Shisi, Kenshin killed many for the hopes of bringing about a new era filled with peace. His name was changed to Kenshin by his master which means "heart of sword." After the revolution, Kenshin vows to never kill again and sets off as a rurouni(wanderer) with a sokabato (or sakaba:reverse blade sword).
Misao meets up with Kenshin on his way to Kyoto when she steals some money from bandits. She follows Himura on his way thinking that he knows the whereabouts of her clan, The Oniwaban Group.
Megumi was forced to use her medical skills to produce opium for Kanryu, a druglord, and madman. However, Kenshin, Sanosuke, and yes, Yahiko come to save the day.
Sanosuke was once a fighter for hire nicknamed Zanza. He was hired to kill Kenshin and ended up getting his butt kicked. Sano decided to drop the ganster life and remain friends with Kenshin.
Leader of the third squad of the Shinsengumi during the revolution, Saito Hajime, also known as the wolf of Mibu, was a skilled swordsman. Although he fought on opposite sides of Kenshin, they were much alike. In Kyoto, Saito even joins forces with Kenshin to try and defeat Makoto Shishio.
Aoshi become leader of the Oniwabashu when he was only 15 years old. The Oniwaban Group were the protectors of Edo castle during the revolution, and were the bodygaurds of Kanryu. However, Kenshin defeated Aoshi to retrieve Megumi and the rest of the Oniwabanshu were killed by Kanryu. Aoshi is not seen again until the Kyoto Arc.
Kenshin had encountered Yahiko when he found him pick-pocketing a man, and then returning the money. They found out that he was an orphan and was forced to steal. Kenshin, however, saved him from this lifestyle and was taken into the dojo to the the Kamiya Kasshin style.
The successor of Battousai the manslayer. He took over the role of the Manslayer in the Shadows in Kenshin's absence.But, when the government wanted to get rid of him, they set him afire. Now in the peaceful Meiji Era, he rises from the shadows to try and take over Japan.
Kenshin Himura's master. He first met Kenshin, then known as Shinta, when he saved him from being murdered. He taught Kenshin until he was 14, when he had decided to leave and fight in the revolution with the Ishin Shishi.
Seta is an assassin with a smile always on his face, even when he's killing. His inability to show any other emotions but joy were caused during his childhood, when life was so difficult all he could do was smile.
Makoto Shishios underground mafia scattered across Japan. They are called to Kyoto when Makoto tries to overthrow the Meiji government.
