AGROBACTERIUM-MEDIATED TRANSFORMATION ON
MONOCOTYLEDONOUS PLANTS AND ITS PROSPECT ON SUGARCANE

ABSTRACT

Agrobacterium-mediated transformation that has been well established on dicotyledonous plants, is also being developed on monocotyledonous plants recently. To date, several monocotyledonous plants that are, Liliaceae, Amaryllidaceae, Iridaceae, Dioscorea and Graminae especially on rice and maize, have been reported to be transformable by Agrobacterium tumefasciens. Agrobacterium-mediated gene transfer has advantages over other methods including the high efficiency of transformation, the simplicity of procedure and the economical value. In order to obtain the success and high efficiency of transformation by Agrobacterium on monocotyledonous plants, various factors have to be considered, such as the age and type of plant tissues, strain or the opine type of Agrobacterium, the addition of phenolic compound (eg. acetosyringone), and the using of super virulent vectors. Until now, the developing of genetic transformation method on sugarcane is only focused on direct gene transfer. Considering the success of Agrobacterium-mediated transformation on several monocotyledonous plants as mentioned before, this method is potential to be applied on sugarcane to improve the genetic engineering program of high yield sugarcane cultivar.
