     A wide variety of spectroscopic methods have been adapted to investigations of biochemical interest. Applications range from those which are pragmatic and straightforward (for example, the measurement of protein or nucleic acid concentration by measurement of the absorbance of ultraviolet radiation) to those which are esoteric and complex (for example, the solution of the three-dimensional structure of proteins and nucleic acids using X-ray diffraction methods or two-dimensional nuclear magnetic resonance methods).

SLM 4800 C Florescence Spectroscopy Systems Top
The 4800 C combines the capabilities of a sophisticated steady-state spectrofluorometer with the additional features of subnanoscond lifetime instrument. Steady-state operations includes obtaining excitation and emission spectra, polarization, and fluorescence intensity measurements. The dynamic capabilities of the instrument include the measurement of fluorescencent lifetimes and phase/modulation spectroscopy based on available operating frequencies of 6, 18, and 30MHz. Phase-resolved spectrofluorometry combines the steady-state and dynamic capabilities of the 4800 C to yield new capabilities in subnanosecond fluorscence spectral analysis. The instrument is equipped with a supplementary detection channel facilitating data acquisition. The entire system is computer controlled permitting a variety of data acquisition modes and affording capabilities in data manipulation.

The instrument has recently been upgraded to permit greater flexibility in data acquisition. Available accessories permit a wide variety of infra-red experiments including a diffused reflectance, attenuated total reflectance, and transmittance modes of operation.

Jasco J-500 A Spectropolarimeter (Circular Dichroism) Top
The instrument has recently been upgraded to permit reliable data acquisition in the far UV to a wavelength of 180nm. Data Acquisition as well as instrument parameter control has been computer interfaced with an IBM PC XT. Hard copies are generated on and HP 7475A Printer/Plotter. A precision Haake circulator is available for use with thermostatted cells permitting determination of temperature dependent alterations in structure. Circular diachronic spectra in the region of peptide bond absorbency (180nm-240nm) can provide a measure of protein secondary structure on deconvolution of the ellipticity envelope. A program written by Kathleen Williams of the department staff is available for this aspect of data analysis.

IBM AF 270 MHz Fourier Transform Nuclear Magnetic Resonance Spectrometer Top
Probe heads for the detection of  1H/13C (10mm), 19F (5mm), and a tunable probe (10mm) are available permitting acquisition of spectra of all magnetic nuclei including  31P, 15N, etc. This instrument is of Bruker manufacture with and Aspect 3000 computer system and resident software. The system is capable of a variety of experimental operations including the acquisition of two-dimensional spectra. While the instrument is adequate for many classes of biological NMR experiments, a departmental priority is the acquisition of a high field (500MHz) instrument, which will permit extension of the types of magnetic resonance investigations.
