Working
in Environmental Microbiology and Molecular Ecology Laboratory at department of
Microbiology, University of Dhaka in collaboration with Department of
Microbiology, University of Bergen, Norway, in water pollution projects (NUFU,
Norway). The title of the thesis is “Isolation, Identification and molecular
characterization of Shigella from
water and food in slum areas in Bangladesh”.
Short Description of the methods: Sample collection and transport. Search of presence of Shigella in sample: by PCR, fluorescent microscopy. Viability can be checked by using INT /AODC/ CTC/ Nalidixic acid; Resuscitation of injured Shigella cells: for above purposes design of special pre-enrichment and enrichment broth. Confirmation of Shigella : by biochemical test (e.g.: API 16), PCR amplification with ipaH, ipaBCD, stx primer, agarose gel run and band analysis with ‘Gel Doc’ machine with computer software, colony blot hybridization with non-radioactive labeled complementary DNA, ELISA (using Shigella specific monoclonal antibody), Southern blot transfer. DGGE; Characterization of isolated Shigella: protein profile analysis by SDS PAGE, antibiotic sensitivity pattern, relationship of plasmids profile with antibiotic sensitivity pattern. Community structure analysis, FISH, DAPI. Ribotypping.
