SvS's Biology: Plant Epidermal Layer & Stomates

This technique may assist in Area 1,3,4 and 5 of the Work Group tasks...

Aim:To remove and extract the epidermal layer of a plant leaf for viewing under a microscope.

Hypothesis:The lower epidermis of a leaf will have a high number of stomates.

Equipment:
         
Plant leaves
                Clear nail varnish
                forceps
                 microscope slide and coverslip
                microscope

Procedure:
                paint a layer of clear varnish over a leaf lower epidermis
                allow to dry and then repeat
                use forceps to tear and then peel away a section of the varnish
                varnish should have epidermal layer attached
                place section on microscope slide, add 2 drops of distilled water
                position coverslip and view under 100x then 400x magnification
