First...the sample

You might all remember an episode where I analyzed some DNA using a Southern blot. Well, I'm here today to explain how it's done. It takes a bit more time than portrayed on the show, but it's a straighforward procedure I think you'll all understand.

So, let's get started. To save time, we will start with our DNA sample already prepared. You can use DNA from blood or tissue, or you can use a sample generated by PCR.

PCR stands for polymerase chain reaction which is a method to amplify DNA so that you can see results from a very small quantity. Remember those commercials where the girl told her friends about the shampoo and they told two friends and they told two friends? Well, that's how quickly your sample is amplified.

Once you have a DNA sample, a gel is poured using agarose in a casting tray with a comb to generate sample wells. It is a bit like a slab of jello when you're done.
