1930s New York City. The gambling community is upset by the arrival of a new Salvation Army mission ("Fugue for Tinhorns", "Follow the Fold"). Nathan Detroit runs a nightly floating craps game, but he has two problems. The first is that he needs a thousand dollars to hold his game in the Biltmore Garage ("The Oldest Established"). The second is his fiance of 14 years, Adelaide, who wants him to give up gambling and finally marry her. In order to solve his first problem, he bets Sky Masterson a thousand dollars that Sky can't take one of the mission workers to Havana. At first it looks like an easy win for Nathan ("I'll Know"), who then goes to take care of his second problem at the Hot Box Cafe ("A Bushel and a Peck") and promises to elope to Maryland with her that night ("Adelaide's Lament"). Nathan's friends are worried about his divided priorities ("Guys and Dolls"), especially since Big Julie from Chicago just showed up hoping for big money at the craps game. But Sky managed to get Sara to Havana, getting her quite drunk in the process ("If I Were a Bell", "My Time of Day", "I've Never Been in Love Before"), so his money from losing the bet naturally never comes and so meanwhile the craps game is taking place in a sewer, and Big Julie won't let it end until he wins, which he isn't, so Adelaide is still waiting for Nathan ("Take Back Your Mink"). Sara's coworker tells her not to give up on Sky ("More I Cannot Wish You"), which she had been about to do. The sewer craps game takes a turn when Sky bets all the other players on a single roll. If they win, he owes them each a thousand dollars; if he loses, they all have to come to a late-night service at the mission, as Sky had promised Sara, since the mission had been hard up for customers and was at risk of being closed down ("Luck be a Lady Tonight"). Naturally, he wins, meaning that Nathan still can't elope with Adelaide ("Sue Me"), but the gamblers all show up for services ("Sit Down, You're Rocking the Boat"), saving the mission as well as both relationships ("Marry the Man Today"), and they all live happily ever after.
