We will start assuming you got to Mérida city by bus and you are at "Terminal de Pasajeros" (bus station). In front of station lies Las Américas Avenue, and you should get on a bus or van going up, and who gets to "el centro" (downtown). Fare averages Bs. 100 (US$ 0.17).
Mérida city is about 1550 m (5085 ft) high on an inclined mesa, going from southwest (low part) to northeast (high part). It is flanked by two sierras: Sierra Norte (North Sierra) and Sierra Nevada (Snowed Sierra, at south). Downtown is divided in Avenues and Streets. Avenues are numbered from N° 1 through N° 8 and are the longest; the streets are crosswise and span from N° 13 (high part) through N° 38 (low part).
The bus or van you took in front of bus station will take you to 25th street between 2nd and 3rd avenues. You should get to Av. 4 and walk up slope until intersection with 19th St.. At this corner is the bus stop to Tabay town, the initial point for a lot of Sierra Nevada trekking routes. Fare is Bs. 160 (US$ 0.27) and you should add Bs. 20 weekends and holidays. This buses work (officially) from 5:30 am until 10:00 pm. However, it is very hard to find transportation after 8:00 pm. It is a 35 min. ride.
